Do you have to use this switch while the amp is off or on standby?

Nope. It's perfectly safe to change from half to full power with the amp in operation, per DR.Z

I do not know if it matters or not, but I always put mine on standby before I flick the switch.

Either way is safe. I'm really careful with my amps, and being an old retired EE I love circuit analysis, so I do it on all my amps. The Z-Lux is different from other amps with half power switches, and the way Doc did it is really clever, like all his circuits tend to be. Without going into specifics, the power is cut in half but the amp doesn't know it really happened. Maybe Doc can chime in on this, but I'm not going to reveal his intellectual property.
Each amp is different. I always turn my Maz 8 to standby prior to activating the Pentode/Triode switch.

Don't own a Z-Lux, but I've switched the half/full switch on my Remedy many times, on the fly, while in operation....haven't experienced any issues.
