I'm used to a big change in an amp when changing the 12ax7 tube - usually in the preamp stage. does changing the other tubes besides the KT-66 power tubes have a big effect on the Route 66?

I can only speak from personal experience, but I've found that the EF86 can bring a bit more treble to the mix, depending on the tube. It doesn't seem to alter the overdrive characteristics of the amp however.
The 12AX7 (or 5751) can alter how soon the amp breaks up. The hotter the phase inverter, the faster the break-up comes along.
The rectifier tube can also alter when the break-up comes along. 5U4 = earlier. 5AR4 = later (cleaner amp)
KT-66 tubes all have their own characteristics of overdrive. I'm a Golden Lion guy.
