I just ordered some Sperzel locking tuners for my Les Paul Standard. Has anyone installed these on an LP before? Any holes that need to be drilled?
I guess I want to make sure that I will be able to put the stock tuners back on the guitar without any problems or signs of previous modification. Any help is greatly appreciated...

Depends...
When I replaced my crappy Klusons on my 69-Goldtop for real Grover tuners - the holes for the fastenings screws didn't match up. The drilled holes for the shafts were OK.
So, I took took ordinary matches & brooke them off in suitable lengths - pressed them into the the old screw holes with some glue and let dry out properly.
Then I could drill new holes for fastening screws for the Groover tuners.

Yeah, looks like small holes for the guid pins have to be drilled, but that should be all. Hopefully the tuning peg shafts will fit just fine...

I've never done Sperzels on a 3+3 headstock, but the install on a 6-inline headstock was pretty easy. The right size drillbit and a gentle touch are all you need.
Measure out the proper depth on your drillbit and put some tape on it to let you judge hole depth. The locating pins for the Sperzels don't have to be drilled that deep. It's also a good idea to use a small pin punch to exactly set the location you want the hole, so the bit won't "walk" across the wood before it digs in. Better bits won't do this as they have the split point to prevent it (check out the DeWalt bits, but many others also have this feature).
