I know it can take KT-66 (by design) and I read that it will take 6L6s, but will it also run on 6V6 or EL-34s? I am particularly interested in EL-34s, but I am guessing the PT voltages may be wrong for it.

If I remember correctly, you can only use KT-66's.

You need to ask the Doc to be sure on this one before trying it. My guess is 6V6's definitely not, EL-34's...maybe.
But more importantly.... why would you want to? There are 6L6 and EL-34 amps by the score, but very few good KT-66 amps. The Rt. 66 is such an outstanding amp as it is. If the KT-66's aren't doing it for you for some reason I think you'd do better to sell it and buy something else to suit your tone needs.

I know this isn't the best comparison but using 6l6's in a KT45 would be like sticking 8 guage on a strat. It can be done but your tone is going to fall very short of it's true potential. If the good Dr designed the amp around the hi fidelity KT66 what would the point be to stick in 6L6?

Route 66 is specifically designed around the KT-66 tube. It may take something else but I'm doubtful it would sound good. If you'd like more of a EL-34 sound in a Route 66 "package" then KT-45 is the amp for you.
