After I had a tube failure before a gig last week, it made me think that it might not be a bad idea to have a spare set of tubes just for an emergency. Since 6L6's are cheap and easy to get, I thought I may go this route (pun intended.) ;D
So even though the 66 was designed around the KT66, throwing in a pair of matched 6L6's won't do any damage, right?
And of course, I am just talking about an emergency "at the gig KT66 failure" type of situation. I'd go right back to KT's ASAP.

KT66 draw more heater current then the 6L6, 1400 mA vs. 900 mA if I'm correct. That may be an issue for the tube more then the amp ... just speculating.

I think the tubes will only draw what they require so it shouldn't be an issue .I know one guy that runs KT66's in his Fender amp all the time and likes them . Not sure how 6L6's would sound in a route 66 , anyone tried them ?
