Just wondering if there is any difference in tone between a 2x10 combo and a head with 2x10 z-best, also should I have any concerns about buying a used 2x10 z-best cab. Is there any problems i could run into buying it used?

Well sir, I cannot give you an accurate comparison (since I haven't tried a 2X10 combo), but I think I can offer a similar situation.
I've got a Maz38 head and several cabs. I have the 2X10 open back, a 2X12 open back, and a 2X12 Z Best among others.
I would think that that the 2X10 combo would sound fairly close to my setup with the 2X10 open back...
Now the Z Best on the other hand is gonna be a LOT different (more bass, deeper tone, more "perceived" volume, since the cab is more uni-directional).
When I compare my open back 12 with the Z Best 12, that's what I hear.
So - what I'm trying to say is that there will be a perceptable tonal difference between a combo and a head/ZBest setup.
As for you question about buying a used ZBest - not much to worry about. The only possible problem I can think of would be blown speakers due to previous owner's use of a high wattage amp....

Yes, there is a difference in tone between the 2X10 Combo and a head/2X10 cab. I played both extensively before I bought my setup. The combo is definitely brighter than the Head/2X10 rig. I'm buying all heads now so that I can mix and match different cabs and amps, more flexability that way.

There probably will be a difference because the Z-Best cab is closed back with the bass porting. So your bass response is going to be a bit higher with the Z-Best. Also, the Z-Best is going to be tighter as well.
The open back 2x10 is going to use sound reflection out the back and reflect off the wall behind you. Bass response will be less because it's open back. No problem though, the bass pot is totally flexable and can compensate.
As far as used 2x10's? Uh, probably not an issue as long as they were taken care of like any other piece of equipment. On the bright side, they are already broken in. Also, the 10's by DRZ are hand made by Doc along with some guidance from Weber. So they're the best 10's on the market.
