The Dr's website says the M12 has the same front end as the Route 66, so I figure it's another highway... but which one?
- There's an old highway in Michigan, state route 12, which no longer exists - There's a canceled British motorway M12
These don't seem like good candidates.

I'm thinking the M is after doc's first name Mike and the 12 is because it's 12 watts. Just my uneducated guess though. ?

There is also a road between Detroit and Chicago M12 (also kniwn as Michigan Svenue). The two cities have a ton of music history (Chicago Blues and Motown). So it is that road plus the one in the UK Doc was paying tribute.
