Hi,
I have recently bought a Dr Z Route 66.
The owner before me had a 5y3 rectifier in it. I put in a JJ GZ34 (5AR4) in it.
Do I need to re-bias the amp and if so how do i go about doing this? Is it complicated?

Yes you should rebias with the new rectifier in there. You might red plate the output tubes otherwise.
The easiest, safest way to bias would be to let a knowledgeable tech do it. There are tools available to make it easier for when a person lacks a tech's expertise.
I got started with a groove tubes bias tool. It came with a book. They probably don't make them anymore. If you are adventurous Duncan amps and many other sights offer more complicated ways to use a am meter and other tools.

OK. Thanks for your help. I think i will take it into the tech and get a re-bias done.
